I replace the heater core and got all the air pockets out of the system still over heated then replaced the thermostat and still overheats and loosing water/antifreeze. What could be the problem?

The 60° V6 has a problem with the intake manifold gasket failing.
If your oil is not brown like a milkshake then you are lucky that the coolant is getting sucked in to a combustion chamber and just being burnt up.

GM has an updated gasket kit with new bolts for about $65.
