I have a 1995 Toyota 4 runner 160K miles v6 engine we replaced the thermostat, but now when I run the car the car overheats. We tried to replace it again and noticed that the radiator water was hot, but the engine water was cold. What can be the reason. We took off the thermostat and tried it and it is properly working. What else can it be?

You replaced the Stat now its hotter than usual-its fine w/o the Stat. Try a defective new thermostat. Its getting hot because the coolant is not flowing thru the engine, its being block by the Stat.

And who knows might be an air lock in it-try doing below and report back

BLEEDING COOLING SYSTEM:

Always bleed air from cooling system after replacing coolant. Set heater for maximum heat. Remove radiator cap. Loosen drain plug and remove drain bolt (if equipped) from engine block. Drain coolant reservoir. Fill coolant reservoir to MAX mark with 50/50 water-coolant mixture. Loosen bleed bolt and fill radiator up to base of filler neck. Close bleed bolt when coolant flows out without bubbles. Tighten bleed bolt. With radiator cap removed, start and

Ok I am either retarted or just a girl, but where is the drain plug from the engine block. I know where we drain the water from the radiator, but not from the engine.
