My car will not start with the #1 plug in the engine. If the #1 plug is removed it will start. What is causing this?

No, I removed the plug and was checking for spark. When my mechanic friend was checking the compression it did the same thing. I am confused, as I said, "the car will not start with the plug in the engine, what causes this?"
