P0306 code. Missing on cylinder 6. New fuel pump, fuel filter, PCV, new injector spider. New plugs. Good spark when removed, Checked for impulse from ECM. All good.
It still seems like no fuel is getting to the cylinder.

If you're experiencing a persistent misfire on cylinder 6 (P0306 code) in your 2000 Chevrolet Express, despite having replaced the fuel pump, fuel filter, PCV valve, injector spider, and spark plugs, there are a few additional steps you can take to further diagnose and address the issue:

Check Fuel Pressure: Confirm that the fuel pressure is within the manufacturer's specified range for your vehicle. Use a fuel pressure gauge to measure the pressure at the fuel rail. If the fuel pressure is low, there may be an issue with the fuel pressure regulator, a clogged fuel line, or a problem with the fuel pump wiring.

Inspect the Fuel Injector: Even though you replaced the injector spider, it's possible that the injector for cylinder 6 is still not functioning correctly. Consider swapping the injector for cylinder 6 with another cylinder to see if the misfire follows the injector. This will help determine if the injector itself is the problem.

Check for Vacuum Leaks: Vacuum leaks can cause a lean fuel mixture, leading to misfires. Inspect all vacuum lines and connections, including the intake manifold gasket, for any signs of leaks or damage. You can use a smoke machine or a can of carburetor cleaner to assist in locating any vacuum leaks.

Perform a Compression Test: Conduct a compression test on cylinder 6 to check for any issues with compression. Low compression could indicate problems with valves, piston rings, or head gasket leaks, which can contribute to a misfire.

Verify Ignition Spark Quality: While you mentioned that you have good spark, ensure that the spark plug wire or coil for cylinder 6 is providing a strong and consistent spark. A weak or intermittent spark can cause misfires.

If you've performed these additional steps and the misfire issue on cylinder 6 persists, it's recommended to consult a professional mechanic or a Chevrolet dealership for further diagnosis. They have access to specialized diagnostic tools and expertise to pinpoint the exact cause of the misfire and provide the most appropriate solution.
