Someone when I purchased the car just had changed plugs, upon further investigation he put in Autolite XP-not NGK as recommended. Toyota is very specific when it comes to plugs, are autolites the same as NGK and can these wrong plugs cause a misfire PO300 causing the CEL to come on?

I kind of doubt it with the plugs but it may be a coil pak/plug wire problem instead. If you have to replace a coil make sure to use factory coils as aftermarket ones seem to give trouble.
