I recently had problems with battery on 323 diesel turbo holding it's charge, removed battery abnd found there was very little water in cells, not covering plates, so re-filled to just above plates and re-fitted battery after giving it a charge. Stupidly put cables on wrong way round, everything is now dead. Could this be a main fuse? Or is it worse?

Most likely yes, it is a fuse/fuses. Also look for fusible links at the battery that may be burnt through. They look like wire, but will burn through at a prescribed amount of draw, just like a fuse.
