Could be a bad injector or low compression. Do a complete compression test first to be sure you are working with an engine that is in good mechanical shape. You can spray a small stream of water onto the exhaust ports right at the cylinder head with engine running, the one that doesnt burn the water off quickly (coolest) will be the missing cylinder.
