1996 Dodge Caravan Ticking Sound

My husband just put a used engine in this van. We took it to our mechanic and it checked out ok. It ran fine for about a week. Since then it has developed a loud ticking sound that starts a few seconds after being started. My husband thinks the sound is coming from underneath and could be the transmission. It loses transmission fluid, but never enough to see it on the ground. It is shifting hard now, but he thinks that could be because he overfilled the trans. fluid. Any help would be appreciated.
