I have a blazer with the 4.3 li vortec., When I 1st start and drive all is great but about 6 min into driving. The vehicle losses its ability to accelerate if I press the gas down like taking off from a stop light however if I stop and restart it I does run like when I first started the engine for about another 6 min, replaced fuel filter and pump never had this before. What could it be?

Lack of acceleration, chugging, hesitating, bogging etc. Could be caused by one of the following below.

Inspect and test all the following listed below and get back with some results

Oxygen sensor.
Catalytic converter.
Fuel injectors dirty/sticking.
Mass airflow sensor/Airflow meter.
Throttle position sensor.
Manifold absolute pressure sensor.
EGR Valve
Fuel pressure regulator leaking or defective fuel pump.
False air leakage.
Fuel contamination.
Foul/defective spark plugs.
Open spark plug wires.
Ignition coil/Coil packs defective.
Incorrect ignition timing.
Cap and rotor.

Note:If it doesn't apply disregard.
