Can you add black rtv silicon to the back of transmission cooler lines to stop leakage from the fitting?

I don't know why it's leaking in the first place. I'm not there. You just need to repair/replace whatever is damaged. All I can tell you for sure is that silicone will not hold back that fluid.

Ive tried replacing o-rings that doesnt work, is there any type of sealant that can work/be used.

No, it's under pressure. It HAS to be repaired properly.
