1988 Chevy Silverado Odd vibration

The Silverado has a new crate engine installed, with 13,000 miles. There is a vibration that starts when it is in overdrive, and it feels like it bogs the engine down. I checked the U-joints, tires are balanced, and no weights have come off of the driveshaft. When I was checking everything, it looked like the piece that goes onto the output shaft has play in it. The transmission has 74,500 miles on it, and it is a 700-R4Could that play in the back of the transmission be the cause of the vibration? If so, what would need to be fixed?
